PCNA at a glance

PCNA (proliferating cell nuclear antigen) is DNA polymerase delta auxiliary protein acting in homotrimeric form as a core of replisome during DNA synthesis and repair. PCNA is expressed in the nucleus of all proliferating cells. In response to DNA damage, it is ubiquitinated and is involved in the RAD6-dependent DNA repair pathway. PCNA is a useful marker of DNA synthesis, as its form not involved in DNA synthesis degrades in histological preparations in the presence of organic solvents. This usage has not only diagnostic, but also prognostic value. Recently also another approach has been intensively studied. For PCNA interacts with multiple proteins involved in DNA replication, repair, and RNA transcription, as well as with a wide range of other factors related to regulation of these processes, it can be used as a unique key target to counteract in one step the DNA replication, repair and transcription processes. This method is very promising, as it has been adapted specifically for neoplastic cells, leaving healthy tissues unharmed. Intervention against PCNA functions turns out to help also against DNA viruses.

The mouse monoclonal antibody PC10 recognizes human PCNA in Western blotting, flow cytometry, immunoprecipitation, immunocytochemistry, and immunohistochemistry application. It cross-reacts with mouse, rat, non-human primate, chicken and Drosophila PCNA.
